Berline was born in Caldwell, Kansas, started playing the fiddle at age five and quickly developed a talent for it. In 1965, he recorded the album Pickin' and Fiddlin' with the Dillards. He graduated from the University of Oklahoma in 1967 with a teaching degree in Physical Education, and joined the Bluegrass Boys shortly after. Byron Berline first met Bill Monroe in 1965 and was offered a job then. After his discharge, two years later, he joined Dillard & Clark on the album Through the Morning, Through the Night. In 1969, he moved to Southern California. Berline won the National Oldtime Fiddle Contest Championship in 1965 in Weiser, Idaho.

Like his contemporary Vassar Clements, fiddler Byron Berline expanded the sonic possibilities of bluegrass, adding elements of jazz, pop, blues, rock, and traditional country to the genre. In addition to being a popular solo act, he performed as a session musician on a number of albums, including records by the Flying Burrito Brothers, Stephen Stills, the Dillards, Gram Parsons, the Nitty Gritty Dirt Band, Emmylou Harris, Kris Kristofferson, and James Taylor. He remained with Dillard and Clark until 1971, when the group disbanded. While with them, he played sessions for a number of other artists, including the Flying Burrito Brothers' debut album, The Gilded Palace of Sin.

Berline later formed the band Byron Berlin and Sundnace with a debut album release in 1976 and Berline, Crary, and Hickman which was later renamed to California. California was awarded as Instrumental Group of the Year in 1992, 1993 and 1994 by the International Bluegrass Music Association. In 1995, Berline moved to Guthrie, Oklahoma where he opened the fiddle shop Double Stop and formed The Byron Berline Band. In 1997 he founded the annual Oklahoma International Bluegrass Festival
